摘要
目的检测帕金森病(PD)模型猴双侧尾核细胞外液兴奋性氨基酸(EAA)含量的变化,探讨EAA在PD发病机制中的作用。方法应用1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)经一侧颈总动脉注射制备偏侧PD猴模型(共4只),模型制备后3.5年,根据模型猴运动症状及对阿朴吗啡(APO)诱发旋转行为的反应,将模型猴分为模型组(2只)及模型恢复组(2只),采用颅内微透析技术结合反相高效液相色谱-荧光法在体检测猴双侧尾核细胞外液EAA及抑制性氨基酸(IAA)含量。结果模型组中MPTP损毁侧尾核细胞外液谷氨酸(Glu)、天门冬氨酸(Asp)、甘氨酸(Gly)、γ-氨基丁酸(γ-GABA)较未损毁侧分别增加了2.4、7.2、1.3及5.0倍;而在模型恢复组中MPTP损毁侧Glu、Asp、Gly亦较未损毁侧增高,但增加不明显,γ-GABA含量相差不大。结论尾核部位Glu、Asp增高可能与PD猴的运动症状的产生相关,Glu。
Objective To investigate the role of excitatory amino acid (EAA) in pathogenesis of Parkinson's disease by measuring the level of EAA in parkinsonian monkey models. Methods Four monkey models of hemiparkinsonism were established by unilateral intracarotid infusion of 1 methyl 4 phenyl 1,2,3,6 tetrahydropyridine (MPTP). Three and half years later, according to their response to apomorphine, the monkeys were divided into 2 groups, the model group (2 monkeys) and the model reversible group (2 monkeys). The concentrations of EAA and inhibitory amino acid (IAA) in the bilateral caudate nuclei of the monkeys were measured by brain microdialysis combined with reverse phase high performance liquid chromatography (RP HPLC) and fluorometric detection. Results In the model group, the contents of Glu, Asp, Gly and γ GABA in the MPTP treated side were increased by 2 4, 7 2, 1 3 and 5 0 times, respectively, as compared with those in the untreated side. In the model reversible group, the levels of Glu, Asp and Gly in the MPTP treated side were slightly elevated as compared with those in the untreated side, and the content of γ GABA was not enhanced. Conclusions The levels of EAA, Glu and Asp may be related to the symptoms of parkinsonian monkey models. Glu and Asp may participate in the damage of striatal by MPTP.
