动员自体骨髓干细胞对大鼠急性肾小管坏死的治疗作用

摘要 目的:观察联合应用粒细胞集落刺激因子(G-CSF)和干细胞因子(SCF)对急性肾小管坏死的修复作用。方法:160只8~10周龄雄性SD大鼠随机分为4组:空白对照组(A组)、缺血再灌注组(I/R,B组)、I/RG-CSF+SCF治疗组(C组)、G-CSF+SCF对照组(D组)。C、D组于手术后予G-CSF50μg/(kg.日)和SCF200μg/(kg.日),1次/日×5皮下注射,于术后第5、10、17、24、31天每组分别取8只大鼠处死,流式细胞仪检测静脉血CD34+细胞百分比的变化和免疫组化法检测肾组织CD34+细胞表达变化,光镜下观察肾脏组织病理学改变。结果:C组和D组外周血中CD34+细胞百分比于第5天达高峰,与A组、B组相比,差异有显著性(P<0.05);C组肾组织内CD34+细胞较A组、B组也明显增多(P<0.05);术后第5、10、17天C组肾脏病理学改变均明显好于B组(P<0.05),第24天C组肾脏病理学改变恢复正常,而B组仍异常。结论:粒细胞集落刺激因子和干细胞因子联合应用能加速肾小管坏死的修复。
