摘要
采用MS基本培养基,附加不同组合的激素对2个甘蓝型油菜和2个白菜型油菜的不同外植体进行培养,其中特别比较了AgNO3以及水解酪蛋白和谷氨酰胺对芽苗分化的促进作用,以期获得影响芽苗分化的必要因素.结果如下:(1)AgNO3能大幅度地提高愈伤组织的分化频率,特别是与水解酪蛋白和谷氨酰胺结合使用效果更好,其中子叶愈伤组织分化频率提高幅度最大,达35.8%.此外,AgNO3还能提高花药培养的分化频率.(2)甘蓝型油菜较白菜型油菜的再生能力强.前者最佳的激素组合为6-BA+NAA,后者为ZT+IAA.在外植体中,花丝和下胚轴都有较高的分化能力。
The different explants of two B.napus L. varieties and two B.campestris L. varieties are cultured on Murashige and Skoog (MS) medium supplemented with three different combinations of plant hormones,in which special attention is paid to the effect of AgNO 3,hydrolcasein and glutamin on buds differentiation from calli,with the expectation to get the necessary factors that influence buds differentiation.The main results are as following:(1) AgNO 3 can greatly improve the differentiation frequency.Especially better results could be achieved when AgNO 3 is used together with hydrolcasein and glutamin,among which cotyledons get the greatest improvement,35 8%.In addition,AgNO 3 can also improve the differentiation frequency of anther culture.(2) Compared with B.campestris L.,B.napus L. have better regeneration capability.The best combination of hormones of the former is 6 BA+NAA,and that of the latter is ZT+IAA.In explants,filament and hypocoty1 both have higher differentation capability.5% of the filament can regenerate buds from the base directly in the inducing culture.
