文拉法辛和米氮平对改善小鼠学习记忆功能的影响

摘要 目的观察文拉法辛和米氮平对改善小鼠学习记忆功能的影响。方法选用小鼠记忆获得障碍和记忆巩固不良模型,受试小鼠随机分为8组(n=10),分别为空白对照组,模型组,文拉法辛50、30和15mg/kg组,米氮平10、5和2.5mg/kg组。小鼠连续经口灌胃给药3周,最后一次给药1h后分别采用跳台法和避暗法测定动物的记忆功能。结果与模型组相比,文拉法辛50和30mg/kg组及米氮平10和5mg/kg组能显著改善东莨菪碱所致的小鼠记忆获得障碍;文拉法辛50和30mg/kg组以及米氮平10mg/kg组可明显改善亚硝酸钠所致的小鼠记忆巩固不良。结论抗抑郁药文拉法辛和米氮平能改善小鼠由东莨菪碱所致的胆碱能神经功能受损以及亚硝酸钠所致的脑部缺氧性记忆障碍,且呈明显的剂量-效应关系,提示抗抑郁药具有除抗抑郁外的其他药理作用。 Objective To observe the memory improvement of venlafaxine and mirtazapine against memory impaired mice. Methods Memory impaired mice induced by scopolamine or NaNO2 were randomly divided into 8 groups ( control group, model group, venlafaxine 50, 30 and 15 mg/kg groups, mirtazapine 10, 5 and 2.5 mg/kg groups, n=10). Mice were treated with drugs for 3 weeks. Step down test and step through test were used to observe the learning and memory ability one hour after the last drug administration. Results Compared with model group, the memory ability of memory impaired mice induced by scopolamine in the group of venlafaxine 50 and 30 mg/kg as well as those in mirtazapine l0 and 5 rag/ kg improved significantly, and those induced by NaNO2 in the group of venlafaxine 50 and 30 mg/kg and mirtazapine 10 mg/kg improved significantly. Conclusion Venlafaxine and mirtazapine could improve the memory ability in memory impaired mice induced by scopolamine or NaNO2 in a dose-dependent manner. Besides antidepressive effect these two antidepressants might have additional memory improvement effect.
