卵巢上皮性肿瘤DNA倍体分析及P53表达意义

摘要 目的:观察卵巢上皮性肿瘤(EOT)患者核DNA含量、P53蛋白表达情况,研究其与生物学行为之间的关系及临床鉴别诊断的意义。方法:采用流式细胞术对162例卵巢上皮性肿瘤(EOT)石蜡包埋组织进行肿瘤单细胞DNA倍体分析;同时,采用流式细胞术对P53蛋白表达进行检测。结果:53例良性肿瘤DNA倍体检测均为二倍体;43例交界性中检出异倍体11例,二者有显著差异。恶性EOT与交界性EOT和良性EOT比较均有显著差义。P53阳性表达恶性EOT与交界性EOT和良性EOT比较均有显著差义。显著高于后二者(X2=34.719,P<0.001)。结论:P53阳性表达与DNA异倍体密切相关,可能是肿瘤发生的机制之一。 Purpose:To observe the nuclear DNA contents and P53 expression of the patients with epithelial oarian tumors (EOT) and to study the biological behaviors and the clinical significance of the differential diagnosis.Methods:Tumor single-celled DNA ploidy were analyzed by flow cytometry for 162 paraffin specimens of EOT,as well as the expression level of P53.Results:DNA ploidy of 53 cases with benign tumor were diploid.11 cases of heteroploid were detected in 43 specimens of borderline tumor.There were significant differences between aforementioned groups.Malignant EOT compared with borderline EOT and benign EOT were significant different,as well as the positivity of P53.Conclusions:The positive expression of P53 and DNA heteroploid are closely related,and might be one of the mechanisms of oncogenesis.
