中枢cAMP水平对大鼠吗啡戒断症状的影响 被引量:12

摘要 给吗啡依赖大鼠侧脑室注射环磷酸腺苷(cAMP)及其衍生物二丁酰环磷酸腺苷(dbcAMP),以提高大脑外源性cAMP,此时可加重大鼠吗啡戒断症状,并呈明显的剂量效应关系。侧脑室注射裂解cAMP的磷酸二酯酶(PDE)抑制剂茶碱,以提高大脑内源性cAMP时,也可加重大鼠吗啡戒断症状。相反,侧脑室注射PDE激动剂咪唑,使大脑内源性cAMP减少,则可使大鼠吗啡戒断症状减轻。上述结果可能有助于发展治疗吗啡戒断症状的新药。 The results of the present study showed that the morphine withdrawal syndromes were agravated by intracerebroventricular(icv)injection of exogenous cAMP and its derivative dbcAMP dose dependently in rats.Theophylline or imidazole,as an inhibitor or an activator of phosphodiesterase(PDE)was injected icv to increase or decrease the endogenous level of cAMP in rat brain.The morphine withdrawal syndromes were significantly augmented by theophyline, but apparently relieved by imidazole. The results of our study may be useful to select new therapeutic medicine for morphine withdrawal syndromes.
