摘要
目的检测细胞骨架蛋白Tagln2 mRNA在兔早期妊娠子宫中的表达与调节,以确定其是否与兔早期妊娠有关。方法采用原位杂交及Northern blot方法,检测Tagln2在早期妊娠、假孕及激素处理的兔子宫中表达部位及其表达的调控。结果 Northern blot结果表明Tagln2 mRNA高表达于兔早期妊娠的第2、3天及第7、8天的着床位点。原位杂交的结果表明Tagln2 mRNA在早期妊娠子宫中呈时空特异性表达,高表达于第2至4天的子宫上皮及着床期第7、7.25、8天的着床位点,在妊娠第6至8天的非着床位点及假孕第6至8天的子宫中均没有检测到Tagln2 mRNA的表达;在卵巢切除的子宫中,雌二醇及孕酮均可使Tagln2 mRNA表达上调。结论结果表明Tagln2 mRNA高表达于兔着床期的子宫内膜,而且表达受活性胚胎的诱导,并提示雌孕激素可上调其表达。
Objective: To examine the expression and regulation of Tagln2 gene in rabbit uterus during early pregnancy. Methods: We used in situ hybridization and Northern blot to detect the expression of Tagln2 mRNA in the rabbit uterus during early pregnancy, in the pseudopregnant rabbit uterus, and in the uterus of ovariectomized rabbit supplemented with steroidal hormones. Results: Northern blot analysis showed a high level of Tagln2 mRNA expression in the uterus on day 2 to 3 of pregnancy and at the implantation sites on day 7 to 8 of pregnancy. In situ hybridization revealed Tagln2 mRNA expression in a spatio-temporally regulated pattern in early pregnant uterus. Tagln2 mRNA was highly expressed in the luminal epithelium on day 2 to 4 of pregnancy. A low expression level was detected in the glandular epithelium near the myometrium on days 5 and 6. On days 7.0 and 7.25, Tagln2 expression was upregulated at implantation sites and mainly expressed in luminal epithelium and blastocyst at the anti-mesometrial and lateral sides. On day 8, Tagln2 expression was mainly detected in the fusion area and glandular epithelium at anti-mesometrial side, and became stronger in blastocyst and luminal epithelium at mesometrial side. Tagln2 expression was not detected in the inter-implantation sites on day 6 to 8 of pregnancy or in the uterus on day 6 to 8 of peudopregnaney. In the ovariectomized rabbit uteri, Tagln2 expression was up-regulated by either estrogen or progesterone.Conclusions : Our endometrium at the data indicated that Tagln2 was highly expressed in the rabbit peri-implantation plantation sites and its expression was embryo-inducible. The expression of Tagln2 was upregulated by steroid hormones.
