摘要
目的用流式细胞仪(flowcytometer,FCM)检测周围血中CD4+、CD8+淋巴细胞,结合临床情况对HIV感染者和AIDS患者的免疫状况进行评价。方法将抗凝全血进行白细胞分类计数,用双色荧光标记的单克隆抗体染色,经溶血和固定后,用FCM计数,从而得出CD4+、CD8+淋巴细胞数。结果HIV感染者和AIDS患者的CD4+淋巴细胞数都比正常人低,特别是AIDS患者,他们的CD4+淋巴细胞数都低于200个/mm3,临床表现也很差。结论FCM检测结果与临床评价高度一致,而且FCM比一般人工计数法更准确、方便、迅速,同时也证明FCM是监测HIV感染者和AIDS患者的免疫状况的最佳方法。
Objective Usingflowcytometer (FCM) to detect CD 4 +,CD 8 + lymphocytes in peripheral blood, combine with clinical symptoms to evaluate the HIV/AIDS patients′ immune state. Methods Anti coagulated peripheral blood were obtained from 8 HIV AIDS patients and 5 normal persons. After analyzing their total white cell counts, the whole blood was stained with two color immunofluorescence using directly conjugated monoclonal antibody pairs, followed by lysis of red erythrocytes, fixation of lymphocytes, and analysis by flowcytometry. Multiplying the CD 4 and CD 8 percentages by the absolute number of lymphocytes obtained from the total white blood cell (WBC) count divide by the lymphocyte differential percentage, we got the CD 4, CD 8 counts. Results The HIV/AIDS patients′ CD 4 counts were lower than those of normal controls′. Especially, the AIDS patients′ CD 4 counts were all below 200 cells/mm 3 and their clinical symptoms were serious. Conclusion The HIV/AIDS patients' CD 4 counts are highly correlated with their clinical symptoms. The result also shows that FCM is accurate, sensitive and reliable in CD 4 count.
