摘要
以三种柚子(Citrus grandis L.)品种叶片为外植体诱导愈伤组织,以幼嫩茎段诱导不定芽和不定根。结果表明:1.0 mg/L2,4-D与6%蔗糖处理的叶片愈伤组织诱导率最高,最高诱导率达82.5%;6-BA0.5 mg/L和NAA0~0.5 mg/L激素配比诱导丛生芽效果最高,最高可在每个茎段上诱导4个不定芽;1/4 MS+IBA 2.0 mg/L诱导生根效果最好,最高达40%,虽然根系很弱,难以成苗,但可以用于试管微嫁接。所有的培养中,?溪蜜柚和沙田柚的诱导率接近,都比江津柚高大约30%~50%。
Enhanced axillary branching and adventitious root formation from stems,and callus induction from leaves of three Citrus grandis cultivars were studied.The results showed that the leaf callus induction was obtained through culturing on MS + 2,4-D 1.0 mg/L + sucrose 6%,and the induction ratio reached 82.5%;Best multiple shoot induction was obtained through culturing on MS + 6-BA 0.5 mg/L + NAA 0~0.5 mg/L,and reached about 4 buds per stem explant;Best multiple root induction was obtained through culturing on 1/4 MS + IBA 2.0 mg/L,and the induction ratio reached 40%,although the shoots were too weak to develop whole plantlet,and they could be used for micro-graft.The Shatian-Yu cultivar and the Guanxi-Miyu cultivar have similar higher induction ratios,and were about 1.3~1.5 times of the ratios of Jiangjin-Yu cultivar.
