hIGF-1转基因促周围神经再生和抑制肌肉萎缩的实验研究 被引量:2

摘要 目的了解周围神经损伤后人胰岛素样生长因子-1(hIGF-1)体内转基因促周围神经再生和抑制肌肉萎缩的作用。方法构建hIGF-1的真核细胞表达质粒,90只雄性Wistar大鼠,建立坐骨神经再生室动物模型后随机分为3组。hIGF-1治疗组:挤压伤处神经外膜下即刻注射pcDNAhIGF-1和LipfectAmine混合液10μl(hIGF-1 DNA为4μg);模型组注射pcDNA3.1、LipfectAmine和生理盐水混合液10μl;空白对照组:不注射任何物质。根据不同的时间点(2天,7天,14天和28天)观察hIGF-1质粒转染、坐骨神经再生、骨骼肌萎缩、脊髓运动神经元细胞病理变化。结果不同时相再生神经纤维质和量、骨骼肌萎缩程度、中枢神经元的变化等hIGF-治疗组明显大于模型组和空白对照组(P<0.01)。超微结构见hIGF-1治疗组的再生神经纤维成熟度优于其它两组。结论周围神经损伤后hIGF-1体内转基因能促进周围神经的再生和抑制骨骼肌的萎缩。 Objective To investigate the function of human insulin like growth factor -1 (hIGF-1 )gene transfer in vivo to promote the peripheral nerve regeneration and inhibit the myoatrophy. Methods Constructed hIGF-1 eukaryotic cell expression plasmid; 90 male Wistar rats were divided into three groups randomly after the construction of sciatic nerve regeneration chamber model. IGF-1 treatment group:injected the mixture of pcDNAhIGF-1 and LipfectAmine 10μl (hIGF-1 4μg) instantly under the epineurium on the crush point:model group:injected the mixture of pcDNA3.1, LipfeetAmine and physiological saline 10/d: control group:no injection. observe the hIGF-1 plasmid transfection, sciatic nerve regeneration, myoatrophy and the pathological change of spinal motor neuron cell on 2,7.14,28 day. Results hIGF-1 treatment group got better outcome than model and control group according to the test of the quality and quantity of regenerated nerve fiber, the degree of myoatrophy and the change of central neuron( P 〈 0.01 ). Conclusion hIGF-1gene transfer in vivo could promote the peripheral nerve regeneration and inhibit the muscle atrophy after the peripheral nerve injury.
