棉花水孔蛋白基因的克隆及序列分析 被引量:1

Cloning and Sequence Analysis of Gossypium hirsutum Aquaporin Gene

摘要 采用RT-PCR的方法,根据GenBank上公布的棉花水孔蛋白基因序列设计引物,克隆得到4个棉花质膜水孔蛋白基因,分别为GhPIP1;1、GhPIP1;2、GhPIP2;1和GhPIP2;2,测序结果与公布序列的相似性都在98%以上,氨基酸序列在99%以上。这是首次从新陆早系列棉花(Gossypium hirsutum)的叶片中克隆得到,为以后研究棉花的质膜水孔蛋白提供参考。 Adopting the method of RT-PCR and according to the aquaporin gene' s primer of Gossypium hirsutum published on the GenBank,four Gossypium hirsutum plasma membrane aquaporin genes were obtained by cloning,including GhPIP1;1, GhPIP1 ;2, Gh- PIP2;1 and GhPIP2;2. The testing sequence results show that the similarity between the sequences of these four genes and the pub- lished four is over 98% ,and the similarity between the sequences of amino acid is over 99%. This is the first time that we have ob- tained these genes by cloning from the leaves of Xinluzao series Gossypium hirsutum, which supplies a base for the further study about the Gossypium hirsutum plasma membrane aquaporin,and also could provide reference for how to improve the Gossypiurn hlrsutum' s ability of combating drought.
