摘要
库尔勒香梨多糖的粗提物经醇沉后,用正丁醇、20%三氯乙酸以及正丁醇-20%三氯乙酸法去蛋白,过DEAE-52柱层析纯化后,HPLC和Sephadex G-100凝胶柱层析证实为单一组分,并对多糖的单一组分清除自由基能力进行了研究。结果表明:经醇沉后的多糖采用正丁醇-20%三氯乙酸法去蛋白效果最佳,其多糖损失率为34.85%,蛋白质去除率为13.75%。香梨多糖对超氧自由基和羟自由基的抑制效果较好,与其浓度呈线性关系。
The extracting of polysaccharides from Pyrus bretschneideri Rehd were precipitated by ethanol.Polysaccharide is purified by n-butanol,20 %trichloroacetic,n-butanol-20 % trichloroacetic method to remove protein,and then followed by DEAE-52 column chromatography.Polysaccharide is shown to be a single homogeneous component by HPLC and SephadexG-100 column chromatography.the antioxidative activities of polysaccharides is studied.The results showed that it is the best way to remove protein from Polysaccharide using n-butanol-20 % trichloroacetic method,the polysaccharides losing rate is 34.85 % and the protein removed rate is 13.75 % .Pyrus bretschneideri Rehd polysaccharides had strong radical scavenging activity towards·OH and O2-·.The capacity of scavenging free radicals of Pyrus bretschneideri Rehd polysaccharides showed linear connections with the concentration.
