摘要
应用聚合酶链式反应(PCR) 技术, 对3 个杂交稻组合及其相应亲本的核DNA 进行了分析, 筛选出9 个随机扩增多态性DNA (RAPD) 随机引物和1 对简单重复序列(SSR) 引物, 可对供试材料进行真伪和纯度鉴定。该技术具有快速、简便、准确的特点, 每人每天可测定180 份样品, 适用于杂交稻种子的商业化鉴定。
DNA from three hybrid rice combinations(each comprising the male sterile,the restorer and the hybrid F 1)has been extracted and amplified by PCR with different random DNA(RAPD) primers and simple sequence repeat(SSR) primers.Of the 300 RAPD primers and 50 SSR primers,9 RAPD primers and 1 SSR primer were polymorphic in three families,and amplified bands are reproducible.Thus,PCR based DNA analysis is proposed as a convenient tool for the identification of hybrid seed.
