果糖及葡萄糖对精子运动能力存活率及顶体酶活性的影响 被引量:1

Effects of fructose and glucose on the movement,survival and acrosin activity of human sperm

摘要 目的探讨果糖及葡萄糖对人类精子运动能力、存活率及顶体酶活性的影响。方法选取2008年7月~2009年1月在浙江省舟山市妇幼保健院生殖中心接受精液质量检查的男性患者312人,经精液常规分析符合《人类精液及精子-宫颈黏液相互作用实验室检验手册》规定的正常标准者;以无糖的人类输卵管液(HTF)为对照组(n=312),添加果糖成分的研究组1(n=312),添加葡萄糖成分的研究组2(n=312),观察不同配方精子培养液培养前、培养24,48,72 h后对精子运动能力、存活率及顶体酶活性的影响。统计学处理采用SPSS 10.0统计学软件,数据均衡性检验后用t检验或单因素方差分析方法 ,P〈0.05为差异有统计学意义。结果与对照组相比,培养前各项指标差异无统计学意义;培养24,48 h及72 h后,研究组精子的各项指标明显较好(P〈0.05)。两研究组之间相比,培养前及培养24h的各项指标差异无统计学意义;但培养48 h及72 h后,研究组1的各项指标都优于研究组2,但差异无统计学意义。结论精子培养液内添加糖分是必须的,在精子培养24 h之前,添加果糖和添加葡萄糖对精子的运动能力、存活率及顶体酶活性没有明显差别,但培养超过24 h后,以果糖作为精子能量供应物质优于葡萄糖。 Objective To investigate the effects of fructose and glucose on the movement,survival rate and acrosin activity of the human sperm.Methods The sperm quality was detected in 312 male patients in the Center of Reproductive Medicine of Zhoushan Maternity and Infant Health Hospital from July 2008 to January 2009.All the semen specimens used in this experiments met the normal criterion of Laboratory Manual for Examination Human Semen and Sperm-cervical Mucus Interaction.Human Tubal fluid(HTF)without glucose was used as a control group(n=312).HTF with fructose or glucose were used as a test group 1(n=312) and a test group 2(n=312),respectively.The movement,survival rate and acrosin activity of human sperm were detected before and after 24 h,48 h and 72 h of culture with HTF medium containing different supplements.Results Compared with the control group,no significant differences were found in the test groups before culture.After 24 h,48 h and 72 h of culture,all indexes were better in the test groups than in the control group(P0.05).There were no significant differences in all parameters between two test groups before and after 24 h of culture;After 48 h and 72 h,all indexes were significantly better in test 1 group than in test 2 group(P0.05).Conclusion Sugar is necessary for the sperm culture medium.Before 24 h of culture,there were no significant differences in the movement,survival rate and acrosin activity of the human sperm in different HTF medium containing fructose or glucose.However,after culture for more than 24 h,fructose is superior to glucose in improving the sperm movement,survival rate and acrosin activity.
