摘要
阿尔茨海默病(Alzheimer's disease,AD)是老年痴呆症中最常见的类型,其发病机制十分复杂,β淀粉样蛋白(amyloid-β,Aβ)被认为是诱发AD发生和发展的关键因素。一般认为,细胞外Aβ沉积在AD的发病中发挥重要作用,但越来越多的证据表明细胞内Aβ也参与AD的发病过程。研究发现,细胞内Aβ沉积可导致线粒体损伤、内质网应激、钙失衡、氧化应激、细胞凋亡、突触障碍,并可影响细胞信号转导。
Alzheimer's disease(AD) is the most common pattern of senile dementia and its molecular pathogenesis is complicated.Amyloid-β peptides play a pivotal role in the pathogenesis of Alzheimer's disease.The deposition of extracellular Aβ has been suggested to be the cause of AD,but emerging evidence indicates that intracellular Aβ also plays a pathophysiologi-cal role in AD.Possible mechanisms have been suggested to account for the intracellular Aβ toxicity,including mitochondria dysfunction,endoplasmic reticulum stress,calcium dysregulation,oxidative stress,apoptosis,synaptic dysfunction and the inhibition of signaling pathway.
