摘要
观察柴胡有效成分柴胡皂甙对原代培养贮脂细胞(FSC)激活及合成细胞外基质(ECM)的作用。结果:治疗组库普弗细胞条件培养基(TKCncm)、治疗CCl4损伤库普弗细胞条件培养基(TKCtcm)、TKCncm加肝细胞条件培养基(PCncm)组FSC内结蛋白阳性反应明显减弱,对细胞表型转化的形态学特征有一定改善,各治疗组FSC的DNA合成及3H-脯氨酸掺入量均呈不同程度降低,FSC内Ⅰ型胶原含量明显减少。说明柴胡皂甙对肝细胞具有保护作用,直接抑制FSC内DNA的合成,还可抑制FSC的激活,从而抑制FSC合成ECM的能力。
Objective: to observe the effects of Saikosaponin, an effective component of Bupleurum, on activating the primary cultured Fat Store Cells (FSC) and synthesizing extracellular matrix (ECM). Method: utilizing ultrastructure, immunocytochemistry and image pattern analysis to examine contents of collagen type 1, 3, 4 and fibronectin (FN) in FSC; using flow Cytomater to determine content of DNA and measure 3H-Proline incorporation volume so as to detect collagen protein volume in FSC. Result: positive reaction of the desmin was markedly weakened in the groups of TKCnem, TKCtem and TKCnem plus Pcnem. For the transformation of cell phenotype, the morphological character was improved. In other treatment groups on FSC, DNA synthesis and 3H-Proline incorporation volume were decreased at different levels. In all treatment groups, volume of collagen type 1 was decreased markedly, especially groups of TKCnem and TKCtem. Conclusion: Saikosaponin can protect the hepatocytes. It could inhibit the DNA synthesis and the FSC activation so to inhibit the ECM synthesis.
