新生鼠真皮细胞的分离培养及其毛囊诱导能力的鉴定 被引量:1

摘要 目的成功地体外分离培养新生鼠的真皮细胞,为毛囊组织工程提供种子细胞。方法应用中性蛋白酶和胶原酶消化法分离新生鼠皮肤的真皮细胞,进行体外培养后观察细胞的形态变化,并采用细胞免疫组织化学方法及细胞体外移植法检测其毛囊的诱导能力。结果新生鼠的真皮细胞体外培养生长状况良好,呈成纤维细胞样,CD133、多能聚糖表达呈阳性。结论新生鼠真皮细胞体外培养生长稳定,具有较强的毛囊诱导能力,可为其在毛囊组织工程中的应用奠定基础。 Objective To obtain dermal cell from newborn rats successfully which were supplied for the hair follicle tissue engineering as seed cells. Methods Dermal cells were isolated from newborn rats with proteinase neutral and collagenase type I digestion, and then their cellular morphologic changes were observed after cultured in vitro. The hair follicle inducibility was detected with immunohistochemistry and transplantation in vitro. Results The morphology of dermal cell survived well in vitro was identical to fibroblast, and the expression of CD133 and versican were positive. Conclusion Dermal cell of newborn rats can be used as seed cells for tissue engineering due to the simple isolation and stable and active growth.
