摘要
目的研究雌激素及拮抗剂三苯氧胺对子宫内膜癌细胞HEC-1B生长的影响。方法体外培养中分化子宫内膜癌细胞HEC-1B,按实验目的分别加入17β-雌二醇(E2,10-8M)、三苯氧胺(TAM,10-7M)刺激24、48、72、96h后,采用四甲基偶氮唑蓝(MTT)比色方法比较各个条件下的细胞数目。用流式细胞术方法检测加入E2、TAM后的细胞周期变化,比较各个条件下的S期细胞比率(SPF)。结果 MTT结果 :E2、TAM在作用72h和96h后,可显著刺激HEC-1B细胞生长(P<0.05)。HEC-1B细胞加入E2刺激后,SPF在加药72h和96h均增高。TAM作用细胞后,SPF值在药物作用48h起均高于对照组。结论雌激素、三苯氧胺均对子宫内膜癌HEC-1B细胞有促生长作用。
Objective To investigate the effect of estrogen and tamoxifen on the growth of endometrial carcinoma cell line - HEC-1B. Methods The endometrial cancer cell line-HEC-1B was cultured in vitro. The cells were stimulated with 1713-estrodiol( 10-8M )and tamoxifen (10-7M) for 24 h,48 h,72 h, 96 h respectively. The proliferation capacity of the cells was evaluated by means of mircocutrue tetrazolium assay (MTT) and the changes of cell cycle after E2 and TAM introduction were detected by flow cytometry (FCM). The cells percentage in S phase(SPF) in different conditions was compared. Results Both 17β-estrodiol and tamoxifen could significantly stimulate the growth of HEC-1B cell 72h and 96h after E2 ,TAM introduction. HEC-1B cells could increase SPF 72h and 96h after E2 stimulation. The SPF of HEC-1B cell 48h,72h,96h after TAM introduction was higher than that of control group. Conclusion Both estrogen and tamoxifen can stimulate the growth of HEC-1B cell line.
