摘要
目的:探讨组织中糖胺多糖与肿瘤发生的关系。方法:应用自行提纯的硫酸软骨素裂解酶和高效液相色谱(HPLC)二糖分析方法,从细胞外基质代谢的疾病生化学角度,对肺癌患者的病变组织及体液糖胺多糖(GAG)进行了组分和性质的研究。结果:表明肺癌患者组织GAG总量明显高于癌旁的非癌组织;肺癌组织GAG表现为高硫酸化。肺癌患者尿GAG排泄量降低,GAG组分呈严重低硫酸化,GAG的二糖中非硫酸化二糖(△Di0S)含量高于健康对照组15倍。结论:细胞外基质GAG并非仅起单纯的支持和保护作用,还与细胞分化、增殖及其代谢调节等多种生理功能有关。GAG代谢异常与肿瘤的发生。
Objective:In order to understand the metabolism of carcinomas.Methods:GAG contents and composition of lung cancer from 5 patients were studied.Results:When the GAG quantity in neoplastic tissues was compared with that in the normal appearing tissues surrounding the neoplasms;tatal GAG contents ranged from 1.04 to 5 times the normal appearing tissues surrounding the carcinomas.Sulfated dissacharides increased in the GAG of neoplastic tissues.The GAG of neoplastic tissues were oversulfated.Total GAG of unrine from neoplastic patients reduced.△ Di 0S content increased about 15 times in urinary GAG of the neoplastic patients.The GAG was serious undersulfated.Conclusion:The results demonstrated that the abnormal metabolism of GAG was closely related to origination and development of neoplasm.
