大蒜细胞核的分离和核DNA部分特性的研究

摘要 用酶解和机械破碎的方法,经不连续蔗糖梯度离心,从大蒜幼苗组织中分离得到纯净、完整的细胞核。经破膜后,获得纯化的核DNA。热变性分析测得T_m=77.8℃,由此估算核DNA (G+C)%含量为20.73%,是富含A-T型。 Nuclear DNA was isolated from garlic intact nuclei by homogenizing, enzyme lysis,filtration, differential centrifugation and DNase treatments. The purified nuclear DNA wasobtained by sucrose gradient centrifugation. Thermal denaturation measurement gave aT_m value of 77.8℃, corresponding to 20.73% in (G+C)content. Therefore, nuclear DNA be-longs to rich type (A+T).
