摘要
为获得番茄抗根结线虫杂种,以含有温度敏感型抗根结线虫Mi-1基因的高代优良自交系栽培番茄(Lycopersicon esculentum)5678161"为母本,与含有非温度敏感型抗根结线虫Mi-3基因的秘鲁番茄(Lycopersicon peruvianum)LA2823"为父本进行杂交,通过胚挽救技术获得10个胚挽救苗株系;应用序列相关扩增多态性(SRAP)分子标记技术对胚挽救苗进行亲子鉴定,证实杂种的真实性;利用Mi-1和Mi-3基因的特异引物对杂种植株进行分子检测,从杂种中检测出拥有Mi-1基因标记的后代10株,同时拥有Mi-1和Mi-3基因标记的后代1株.
To obtain hybrids resistant to root-knot nematodes of tomato,breeding line "5678161" carrying Mi-1 gene sensitive to temperature was used as the female parent and wild tomato "LA2823" carrying Mi-3 gene insensitive to temperature was used as the male parent.Ten hybrid lines were obtained by embryo rescuing method.Its realities were identified by sequence-related amplified polymorphism (SRAP) method.Ten plants with Mi-1 gene and one plant both with Mi-1 gene and Mi-3 gene were selected out by using primers specific to Mi-1 gene and Mi-3 gene.
