摘要
目的:建立红花地上部分中木犀草苷和木犀草素的含量测定方法。方法:采用HPLC法,色谱柱为Hanbanhedera ODS-2C18柱(5μm,4.6×250mm),流动相为甲醇-0.4%冰醋酸梯度洗脱,检测波长为350nm,柱温30℃。结果:木犀草苷回归方程:Y=59558771X-1755,r=0.999991,在0.0019mg/mL~0.1238mg/mL范围内有良好的线性关系,平均回收率为98.04%,RSD=1.68%(n=9);木犀草素回归方程:Y=76381285X-9981,r=0.999995,在0.0020mg/mL~0.1266mg/mL范围内有良好的线性关系,平均回收率为96.56%,RSD=2.00%(n=9)。结论:该方法简便、快捷、准确、重复性好,可用于木犀草苷和木犀草素的含量测定。
Objective:To determine the content of Luteolin-7-O-glucoside and Luteolin in Overground Portiono of Carthamus tinctorius L. by HPLC. Methods:A Hanban hedera ODS-2 C18(5μm,4.6×250mm) column served as stationary phase and methanol-0.4% glacial acetic acid slution in gradient elution mode as mobile phase; The detection wave length was 350nm and the column temperature was 30℃. Results:A good linearity of Luteolin-7-O-glucoside was in the range of 0.0019mg/mL~0.1238 mg/mL and r = 0.999991; The average recovery rate was 98.04% and RSD = 1.68%(n=9); Luteolin in 0.0020mg/mL~0.1266 mg/mL range has a good linear relationship,r=0.999995; average recovery rate was 96.56%. and RSD= 2.00%(n=9). Conclusion:The method is simple,fast,accurate,reproducible and can be used for determining the content of Luteolin-7-O-glucoside and Luteolin.
