嗜热产甲烷杆菌YT—7菌株的分离和特征

ISOLATION AND CHARACTERIZATION OF A THERMOPHILIC METHANOBACTERIUM YT-7 STRAIN

摘要 采用严格的 Hungate 厌氧技术,从云南省腾冲高温温泉污泥样品中分离一株嗜热自养产甲烷杆菌 YT-7菌株。该菌株细胞呈长杆状,不运动,不形成芽孢,革兰氏染色阳性。在65—70℃下培养时菌体为0.3-0.4×1.5-13μm。分离物仅能利用 H_2/CO_2为能源和碳源,在无机盐液体培养基中生长,细胞最小倍增时间为3.2小时。滚管菌落呈灰白色,半透明,近圆形;落射荧光显微镜检查呈蓝绿色或黄绿色荧光。YT-7菌株的最适生长温度为65—70℃,低于45℃或高于75℃不生长。最适生长 pH 为6.5—7.0。根据形态和生理性状的研究,分离物暂定嗜热自养产甲烷杆菌 YT-7(Methanobacterium thermoautotrophicumYT-7)。 A thermophilic and autotrophic Methanobacterium,designated Metha- nobacterium Strain YT-7,was isolated from the sediment of hot spring in Teng Chong,Yunnan by Hungate roll tube technique.Strain YT-7 was Gram positive,nonmotile,non-sporing and long rods.At 60-70℃,it was long rod (0.3—0.4x 1.5—13um),cell usually occur in single,sometime in chains.The isolate uses only H_2/CO_2 as carbon and energy source.Colo- nies on roll-tube medium are round,translucent,diffuse,tanish-white in color.Fluorescence at 420nm excitation was greenish-blue or greenish- yellow.The optimal temperature for growth was 65 to 70℃.Growth occu- red in the range of pH 6.0 to 8.5,and the optimal growth at pH 7.0. According to its morphological and physiological properties,this met- hanogenic strain is considered to be a member of genus Methanobacterium, to which the name Methanobacterium thermoautotrophicum YT-7 Strain is proposed for the time being.
