脂肪族仲胺在适宜条件下很容易形成强致癌物N-亚硝基化合物,因此研究N-亚硝基化合物及其前身物脂肪族仲胺的灵敏和准确的测定方法是研究癌变过程及其机理的前提条件。 丹酰氯作为荧光衍生试剂已广泛应用于氨基酸、生物胺和多胺的高效液相色谱分析。
A highly sensitive quantitative method for the determination of ultratrace aliphatic secondary amines has been developed.The optimum experimental conditions have been established.Firstly,the sample reacts with dansyl chloride at about 40℃ for 30 minutes and then,the reaction mixtures are injected and separated on C18 column with acetonitrile/water(73/27,V/V) as moble phase,and detected with fluorimetric detection at 530nm as emission wavelength(excitation at 350nm).The calibration curves are linear in the range of 0.055-55 ng for dimethylamine,0.09-180 ng for di-etbylamine,0.12-250 ng for dipropylamine,0.15-320 ng for dibutylamine and 0.19-80 ng for dipentylamine,and the detection limits are 11,30,34,50 and 95pg,respectively.
