摘要
为了研究14-3-3蛋白是否参与了IRF3信号通路,以及14-3-3蛋白质对IRF3转录活性的影响,在真核细胞293T中表达14-3-3蛋白质的三种亚型,并且通过双荧光报告系统研究14-3-3蛋白对TRIF和IKKε激活的IRF3转录活性的影响。结果显示14-3-3蛋白质能够在真核细胞293T中正确表达,过表达14-3-3η能够抑制TRIF和IKKε激活的IRF3转录活性,而且能够直接抑制IRF3转录活性。
In order to study the effect of 14-3-3 protein on the transcriptional activity of IRF3,14-3-3 protein was overexpressed in eukaryocytes 293T,and Dual-Luciferase Reporter Assay System was used to evaluate the effect of 14-3-3 on transcriptional activity of IRF3 stimulated wiht TRIF and IKKε.The results demonstrate that overexpression of 14-3-3η inhibits the activation of IRF3 signaling pathway stimulated with TRIF and IKKε,and can directly inhibit the activity of transcriptional factor IRF3.
