摘要
目的研究瘦素对人单核巨噬-细胞趋化活性的影响,为阐明瘦素在抗感染免疫中的作用提供依据。方法以人单核细胞系为研究对象,采用Boyden小室法检测其趋化活性,采用ELISA测定IL-18和MCP-1的含量变化。结果瘦素处理后的单核巨噬细胞的趋化功能明显增强(P<0.05)。ELISA结果表明瘦素处理后的单核巨噬细胞分泌IL-18和MCP-1的能力也有所增强。结论瘦素可以增强人单核巨噬细胞的趋化功能,这种增强可能与上调IL-18和MCP-1的分泌有关。
Objective To investigate the effects of Leptin on the activity of chemotaxis in mononuclear macrophage. Methods THP-1 cells and macrophages induced from THP-1 were used in this research.The 48-well chemotaxis chamber was used to measure the chemotaxis and ELISA was used to analyze the concentrations of interleukin 8 and monocyte chemoattractant protein 1-α.Results The activity of chemotaxis in mononuclear macrophage was increased significantly after the treatment with Leptin,compared with that before the treatment(P〈0.05).Results Moreover,we found Leptin might affect the chemotaxis by up-regulating the secretion of interleukin-8 and monocyte chemoattractant protein 1-αin mononuclear macrophage
