摘要
目的:探讨bFGF基因修饰骨髓间充质干细胞(Bone Marrow—derived)。方法:96只清洁级成年雄性Sprague—Darley大鼠,随机分为目的基因bFGF修饰BMSCs组(pcDNA3.1-bFGF组)、空载质粒pcDNA3.1修饰BMSCs组(pcDNA3.1组)、骨髓间充质干细胞组(BMSCs组)和DMEM组(DMEM组),每组24只,采用Allen氏脊髓损伤模型,于损伤点注射上述各组相关细胞及培养基DMEM,在注射后1d、7d、14d、21d取损伤脊髓组织,用免疫组织化学及RT-PCR法检测bFGF表达情况。结果:基因修饰组术后各时间点脊髓组织中bFGF表达较空载组和BMSCs组明显增高(P〈0.01),并于7d或14d表达最明显(P〈0.01)。结论:bFGF基因修饰BMSCs移植可以在脊髓损伤模型中表达。
AIM: To investigate the expression of bFGF gene modified Bone Marrow-derived Mesenchymal Stem Cells(BMSCs)transplanting into the injuried spinal cord and expressing bFGF in injuried spinal cord in rats. Methods 96 male adult Sprague-Dawley rats were randomized into 4 groups(24 in each group): pcDNA3.1-bFGF group, pcDNA3.1 group, BMSCs group and DMEM group. Rats injuried spinal cord model was established with Allen' s method. BMSCs were modified by plasmid before the transplantation. After rats model established, 1 × 106 cells modified by pcDNA3.1-bFGF and pcDNA3.1 were injected into the pcDNA3.1-bFGF group rats and pcDNA3.1 group; the same dose of cells were given to the BMSCs group, at the same time DMEM was injected into injuried spinal cord as vehicle groupie. At ld, 7d, 14d and 21d after injection, the bFGF was detected by reverse transcriptase-polymerase chain reaction(RT-PCR)and immunohistochemical method at ld, 7d, 14d and21d after injection. Results: By compared with the pcDNA3.1 group, BMSCs group and DMEM group, the bFGF were better expressed in pcDNA3.1 group at ld, 7d, 14d and 21d after injection(P〈0.01), and the bFGF in injuried spinal cord was significantly upregulated at 7d and 14d after injection too(P〈0.01). Conclusion: Transplantion of gene modified BMSCs can improve the bFGF expression after injection in rats model.
