摘要
目的:探讨超促排卵周期卵泡刺激素受体(FSHR)基因第10外显子Ser680Asn多态性与卵巢反应性的关系。方法:将体外受精(IVF)周期患者136例,按卵巢反应性分组,采用聚合酶链反应-限制性片段长度多态性技术,检测FSHR基因第10外显子Ser680Asn多态性,观察并测定患者临床数据、胚胎实验室数据和妊娠率。结果:①高反应组NS基因型频率显著升高(63.2%,P<0.05)。②SS型患者bFSH水平为(6.81±1.23)IU/L,显著高于NN和NS型。③NS型患者基础窦卵泡数为(13.31±4.33)个,显著多于NN和SS型。NS型患者获卵数明显多于SS型(NS型12.44±5.49,SS型8.41±5.13,P<0.01)。结论:IVF周期卵巢反应性与FSHR基因第10外显子Ser680Asn多态性有关。
Objective: To investigate the correlation of Ser680Asn single nucleotide polymorphism at exon 10 in follicle stimulating hormone receptor(FSHR) gene with the ovarian response of patients treated by assisted reproductive technology in Chinese population.Methods: According to the ovarian response during controlled ovarian hyperstimulation(COH),136 women were divided into three groups: poor response(n=22),normal response(n=57) and high response(n=57).Then FSHR polymorphism was examined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).The clinical and embryological data,as well as the outcomes were compared and analyzed.Results: A significantly increased prevalence of the Asn/Ser variant(63.2%,P〈0.05) was found in the high response group.The FSH level on the 2nd day of menstruation was significantly higher in SS genotype than in genotype NS and NN([6.81±1.23] vs.[5.29±1.15],and vs.[5.74±1.83] IU/L,respectively,both P〈0.05).The number of antral follicles was significantly more in NS genotype than in NN and SS genotypes(13.31±4.33,8.78±2.99,and 9.73±4.16,respectively for NS,NN,and SS groups,both P〈0.01).The number of retrieved oocytes was more in NS genotype than in SS genotype(12.44±5.49 vs.8.41±5.13,P〈0.01).Conclusion: Ser680Asn single nucleotide polymorphism at exon 10 in FSHR gene may play an important role in ovarian response during COH.
