摘要
目的:观察电针任脉对脑缺血大鼠缺血脑区神经干细胞(NSC)增殖的影响,探讨电针任脉治疗缺血性脑损伤的作用机制。方法:采用侧脑室注射逆转录病毒pLXSN-EGFP在体示踪法,将病毒上清注入局灶性脑缺血(MCAO)大鼠侧脑室以标记新增殖细胞,用Nestin免疫荧光染色法鉴定NSC,并计数脑缺血后不同时相缺血侧SVZ、DG区NSC数目。结果:各区不同时相均有新增殖NSC,且电针任脉组增加更明显。结论:电针任脉可明显促进缺血后NSC的增殖。
Objective:To observe the effect of electro-acupuncture(EA)at Ren channel on neural stem cells proliferation of cerebral ischemia rats,explore the mechanism of EA at Ren channel on cerebral ischemic injury.Methods:Retrovirus Plasmid pLXSN-EGFP Trace Technique was adopted.60 male healthy Wistar rats were randomized into model and EA groups,each group was further divided into 7d,14d and 28d subgroups.EA group was electro-acupuncture at points such as 'guanyuan'(RN4),'qihai'(RN6)and 'chengjiang'(RN24)for 10 minutes each day.Each group rats were intraventricular injected Retrovirus Plasmid pLXSN-mediated EGFP after MCAO(modeling)to mark new proliferative cells and Nestin immunofluorescence staining was applied to identify the neural stem cells,and observed thenumber of proliferating neural stem cells at different time.Results:EGFP-positive and EGFP/Nestin-positive were found in dentate gyms(DG)and subventricular zone(SVZ)at different time after cerebral ischemia,which were remarkably increased on the 7th day.The number of EGFP-positive and EGFP/Nestin-positive cells in EA group was more than that of the model group at different time.Conclusion:EA at Ren channel can promote the proliferation of neural stem cells to improve the cerebral ischemic injury.
