细胞周期蛋白A及P21在子宫内膜异位症患者在位和异位内膜组织中的表达 被引量:3

Expressions of Cyclin A and P21 in ectopic and eutopic endometrial tissues of endometrtosis patients

摘要 目的探讨细胞周期蛋白A(CyclinA)及P21在子宫内膜异位症发生、发展中的作用。方法采集子宫内膜异位症患者(观察组)在位内膜(25例)和异位子宫内膜(28例)以及其他良性疾病患者的子宫内膜(对照组35例),用免疫印迹、RT-PCR、免疫组化的方法检测CyclinA和P21表达。结果cyclinAmRNA在异位内膜增生期与分泌期表达量(1.24±0.10)、(1.33±0.21)均明显高于在位内膜(0.72±0.26)、(0.42±0.22)及对照组(0.68±0.09)、(0.35±0.06)(P<0.05)。CyclinA蛋白在异位内膜增生期与分泌期表达量(0.95±0.10)、(0.91±0.18)均明显高于在位内膜(0.53±0.08)、(0.27±0.11)及对照组(0.47±0.11)、(0.22±0.09)(P<0.05)。在位内膜CyclinA表达量与对照组相比均无显著性差异(P>0.05)。P21mRNA在对照组内膜增生期与分泌期表达量(0.21±0.01)、(0.54±0.35)均明显高于在位内膜(0.09±0.06)、(0.28±0.02)及异位内膜(0.06±0.01)、(0.13±0.00)(P<0.05)。P21蛋白在对照组内膜增生期与分泌期表达量(0.74±0.13)、(1.13±0.15)均明显高于在位内膜(0.59±0.21)、(0.62±0.29)及异位内膜(0.40±0.36)、(0.43±0.33)(P<0.05)。CyclinA在对照组内膜增生期表达高于分泌期(P<0.05),P21在对照组内膜分泌期表达高于增生期(P<0.05),而两者在异位内膜表达均失去周期性(P>0.05),两者在异位内膜中表达呈显著负相关(r=-0.738,P<0.01)。结论CyclinA及P21异常表达可能在子宫内膜异位症异位内膜增殖中起重要作用。 Objective To investigate the possible roles of Cyclin A and P21 in the pathogenesis and development of endometriosis.Methods Twenty-five specimens of eutopic endometrium and twenty-eight specimens of ectopic endometrial tissues were obtained from 28 patients with endometriosis (observation group).Thirty-five specimens of endometrium were also obtained from women with other benign diseases (control group).Cyclin A and P21 were detected by Western blotting,RT-PCR and immunohistochemistry.Results The expression level of cyclin A mRNA in ectopic endometrium were (1.24±0.10) at the proliferative phase and (1.33±0.21) at the secretory phase,significantly higher than that in the eutopic endometrium tissues (0.72±0.26,0.42±0.22),control group (0.68±0.09,0.35±0.06) (P〈0.05).Its protein expression was (0.95±0.10) at the proliferative phase,and (0.91±0.18) at the secretory phase,which were significantly higher than those of the eutopic endometrium (0.53±0.08,0.27±0.11),and control group (0.47±0.11,0.22±0.09) (P〈0.05).The expression level of Cyclin A in proliferative phase and secretory phase of the eutopic endometrium had no significant difference compared with those of the control group (P〈0.05).The expression level of P21 mRNA in control group were (0.21±0.01) at the proliferative phase and (0.54±0.35) at the secretory phase,significantly higher than that in the eutopic endometrium tissues (0.09±0.06,0.28±0.02) and in the ectopic endometrium (0.06±0.01,0.13±0.00) (P〈0.05).The expression level of P21 protein in control group were (0.74±0.13) at the proliferative phase and (1.13±0.15) at the secretory phase,significantly higher than that in the eutopic endometrium tissues(0.59±0.21,0.62±0.29)and in the ectopic endometrium(0.40±0.36,0.43±0.33) (P〈0.05).The expression level of Cyclin A in proliferative phase of the control group was significantly higher than secretory phase.The expression level of P21 in secretory phase of the control group was significantly higher than proliferative phase.But the expression level of Cyclin A and P21 in the proliferative phase and secretory phase of the ectopic endometrium had no significant difference (P〈0.05).In the ectopic endometrium they had negatively spearman correlation (r=-0.738,P〈0.01).Conclusion The abnormality expression of Cyclin A and P21 in the ectopic endometrium may be involved in the proliferation of endometrium in endometriosis.
