摘要
目的:观察迷走神经背核(dorsal motor nucleus,DMN)微量注射生长抑素(somatostatin,SS)对大鼠胰腺内神经元c-fos表达的影响。方法:取体质量250~300g健康雄性SD大鼠12只,随机分为两组:实验组(n=6)DMN微量注射100nL浓度为0.4μg/mL的SS,对照组(n=6)DMN微量注射100nL的人工脑脊液(aCSF)。两组均予20%脂肪乳液行十二指肠灌注1h后,取大鼠胰腺标本行c-fos的免疫组化染色。结果:DMN注射SS组大鼠胰腺内c-fos免疫阳性神经纤维阳性面积(6122.5±1410.0vs1875.8±992.0)和阳性指数(0.62±0.02vs0.26±0.07)均明显降低(P<0.01)。讨论:DMN微量注射外源性SS对大鼠胰腺内神经元有显著的抑制效应,说明DMN外源性SS通过抑制胰腺内神经元发挥对胰腺外分泌的抑制作用。
Objective: To observe the effect of microinjection of somatostatin (SS) into the vagal dorsal motor nucleus (DMN) on the expression of c-fos in intrapancreatic neurons.Methods: Twelve SD rats,weighing 250~300 g,were randomly selected and divided into two groups: the experimental group and the control one.100 nL SS (0.4 μg/mL) was microinjected into the DMN in the experimental group(n = 6),while 100 nL artificial cerebrospinal fluid (aCSF) was microinjected into DMN in the control group(n = 6).One hour after the intraduodenal perfusion of 20% intralipid in each group,the rats’pancreases were obtained and studied by immunohistochemistry (IHC) of c-fos.Results: Both c-fos expression positive area and positive intensity of intrapancreatic positiveneurons in the experimental group’s pancreases were significantly decreased after microinjection of exogenous SS into the DMN compared with the control group (6 122.5 ± 1 410.0 vs 1 875.8 ± 992.0,0.62 ±0.02 vs 0.26 ± 0.07,respectively).Conclusion: Microinjection of exogenous SS into the DMN obviously inhibites the activity of intrapancreatic neurons.These results indicate that exogenous SS of the dorsal motor nucleus has an obviously inhibitory effect on pancreatic exocrine secretion by inhibiting the activity of intrapancreatic neurons.
