利用冷冻的GV期卵母细胞制备延边黄牛核移植胚胎的研究

摘要 研究使用玻璃化冷冻、程序化冷冻和超快速冷冻3种方法冷冻延边黄牛的卵母细胞,确定适宜的延边黄牛核移植受体卵母细胞的冷冻方法。结果显示,3种冷冻方法处理的GV期卵母细胞在成熟率上相似,玻璃化冷冻法得到的卵母细胞形态正常率要高于程序化冷冻和超快速冷冻法(P<0.05),超快速冷冻法回收率最低。采用玻璃化冷冻法,核移植重组胚卵裂率明显高于程序化冷冻和超快速冷冻法(P<0.05),在融合率上,3种方法差异不显著。表明玻璃化冷冻方法对延边黄牛GV期卵母细胞冷冻效果优于程序化冷冻和超快速冷冻方法,是一种较适合延边黄牛核移植的卵母细胞冷冻方法。 In order to determine appropriate freezing method for oocyte used in nuclear transfer of Yanbian cattle,three methods were used to freeze oocyte,namely,vitrification,programmed and ultra-rapid freezing.The results showed that maturation rate of oocyte was similar in three treatment groups,morphologically normal rate of oocytes obtained from vitrification freezing method was higher than that from programmed freezing method and ultra-rapid freezing method (P〈0.05),and the recovery rate was lowest in the ultra-rapid freezing method.The cleavage rate of reconstructed embryo was significantly higher in the vitrification freezing method than those in the other freezing methods (P〈0.05),there was no significant difference in the fusion the rate in three methods.It was shown that the vitrification freezing method is better than the programmed freezing and the ultra-rapid freezing method,and it is a more suitable freezing method for freezing oocyte for nuclear transfer in Yanbian cattle.
