摘要
在不同温度下考察脂肪酶的失活动力学,并将失活率不同的脂肪酶使用疏水层析色谱进行分析,查看其保留行为。结果表明,设定40℃时的酶活为100%,则酶活为0~100%在疏水层析柱上表现为2.16~2.39 m in的保留时间区间。但是在该保留时间区间内,脂肪酶的热失活引起的结构变化,在疏水柱上的保留时间并不呈线性变化,使用Z值查看脂肪酶与固定相的结合情况,表明Z值的变化与脂肪酶失活情况下的结构变化不存在明显的对应关系。
Deactivation kinetics of lipase was investigated under different temperature.Lipases with different inactivation rate were analyzed by hydrophobic interaction chromatography and retention behavior was studied.Results show that lipase activity of 0~100% would caused a retention time interval of 2.16~2.39 min when defined lipase activity at 40 ℃ as 100%.In this time interval,structural change caused by heat inactivation and retention time of it was not changes in linear way.When interaction of lipase and stationary phase was investigated by Z value,there was no corresponding relationship between changes of Z and inactivation of lipase observed.
