摘要
通过静态吸附从3种树脂(CAD-40、DM-130、860021)中选择吸附量较大和洗脱效果较好的CAD-40树脂作为纯化甘草酸的最佳树脂,考察动态吸附过程中各因素对CAD-40树脂吸附和洗脱甘草酸的影响。试验结果获得CAD-40树脂纯化甘草酸的最佳条件为:层析柱Ф2×30 cm,装柱径高比为1∶10,上柱液浓度7.0 mg/mL、加样量为5.0 mL,洗脱最佳条件为:洗脱剂乙醇浓度95%,洗脱液流速1.0 mL/min。此条件下洗脱产物甘草酸纯度可达53.3%。
Through the static adsorption experiments from three kinds of resin(CAD-40,DM-130,860021),we chose CAD-40 as the optimal resin for the purification of glycyrrhizin,which has more quantity of adsorbed and better eluting performance.Then we discuss some factors working on the dynamic absorption of the CAD-40 resin,and eventually determined the following optimum condition for absorption of the CAD-40 resin: Chromatography columns Ф2×30 cm,diameter-height ratio with column of 1∶10,glycyrrhizin concentration of 7.0 mg/mL,adding 5.0 mL,and for the optimum eluting condition we chose 95 % ethanol as the eluting solvent and kept the velocity of flow at 1.0 mL/min.As a result,the Purity of the glycyrrhizin product is 53.3 %.
