摘要
目的探讨七福饮对老年性痴呆(AD)模型大鼠学习记忆能力的影响及作用机制。方法将84只SD大鼠随机分为空白组、假手术组、模型组、盐酸多奈哌齐组和七福饮高、中、低剂量组,采用立体定位仪将凝聚状态β-淀粉样蛋白1-42(Aβ1-42)定向注入海马区制备AD动物模型。通过Morris水迷宫的定位航行试验检测各组大鼠学习记忆的能力。采用免疫组织化学SP方法和病理图像定量分析技术检测海马区生长抑素(SS)蛋白的表达水平。结果与假手术组比较,模型组平均逃避潜伏期显著延长(P<0.05),大鼠海马区SS蛋白表达也明显降低(P<0.05);与模型组比较,七福饮各剂量组[2.15、4.30、8.60mg/(kg·d)]平均逃避潜伏期显著缩短(均P<0.05)。与假手术组比较,模型组SS蛋白表达明显降低;与模型组比较,七福饮各剂量组SS蛋白表达均显著升高(P<0.05)。结论七福饮可能通过上调AD模型大鼠海马区SS蛋白的表达,进而改善AD大鼠的学习记忆能力,发挥防治AD的作用。
Objective To observe the effects of Qifuyin on learning and memory ability and expression of somatostatin in hippocampus on Alzheimer’s disease (AD) rats. Methods SD rats were randomly divided into blank control group, sham operation control group, model group, donepezil group and Qifuyin group. The AD model was established by microinjection of Aβ1-42 into the bilateral hippocampus. The learning and memory ability of rats were evaluated with a series of tasks to find a hidden platform in Morris Water Maze. The expression levels of somatostatin in hippocampus were measured by immunohistochemical SP techniques. Results In the place navigation test, the mean escape latencies of the model group was obviously increased compared with sham operation control group (P〈0.05). The mean escape latency of Qifuyin [2.15, 4.3, 8.6 mg/(kg·d)] group was obviously increased compared with the model group (P〈 0.05). The expression of somatostatin in hippocampus of Qifuyin [2.15, 4.30, 8.60 mg/(kg·d)] group was obviously increased compared with sham operation control group and model group. Conclusions Qifuyin has an effect of improving learning and memory ability of AD rats, which is possibly due to the promotion of the expression of somatostatin in hippocampus.
