摘要
目的建立测定熊去氧胆酸血药浓度的方法。方法采用高效液相色谱法联合固相萃取及衍生化法测定血浆中熊去氧胆酸含量,以胆酸为内标物,血浆样品和内标物利用C18固相萃取小柱富集,以2’-溴代-2’-萘乙酮和N,N-二异丙基乙胺为衍生化试剂,衍生化产物利用高效液相色谱法测定。结果熊去氧胆酸在2~60nmol/ml范围内线性关系良好,r=0.999 0。结论本方法准确可靠,灵敏度高,重复性好,可用于血浆中测定熊去氧胆酸的浓度。
Objective To establish a method for the determination of ursodeoxycholic acid in plasma.Methods A solid-phase extraction procedure was developed to enrich ursodeoxycholic acid and cholalic acid,which was the internal standard,and the plasma sample derived by 2-bromo-2'-acetonaphthone and N,N-diisopropylethylamine was determined by high performance liquid chromatography.Results The calibration curve was linear within the range of 2~60nmol/ml for ursodeoxycholic acid(r=0.999 0).Conclusion The method is highly reproducible,reliable and accurate for the determination of ursodeoxycholic acid in plasma.
