摘要
目的建立人肺腺癌的多重抗药细胞系-A549/DDP。方法采用恒定药物浓度,周期性作用的体外诱导法、MTT法、光镜、电镜、活细胞计数法和免疫组化法进行研究。结果A549/DDP对顺铂抗药,对卡铂和鬼臼乙叉甙产生交叉抗药。A549/DDP的细胞形态发生了改变,但其体外细胞群体生长倍增时间与A549的无差异。A549/DDP细胞bc1-2基因表达较A549的增强。结论A549/DDP是抗药性能稳定的多重抗药细胞系,bC1-2基因过度表达可能是其多重抗药的机理之一。
Objective To establish a multidrug resistant human lung adenocarcinoma cell subline-A549/DDP.Methods Pulse treatments by using a single concerntration of Cisplatin in Vitro,MTT method、light microscope、electron microscope、cell counting and immunohistochemical staining.Results A549/DDP showed resistance to Cisplatin (DDP) and also showed cross resistance to Carboplatin (CBP) and Etoposied (Vp-16).The morphological changes were found on A549/DDP cells.The difference in doubling time in vitro between A549/DDP and A549 didn′t possess significance in statistics.Bc1-2 gene expression in A549/DDP cells was higher than in A549 cells.Conclusion A549/DDP cell subline expressed a stable multidrug resistant phenotype.The overexpression of bc1-2 gene in A549/DDP cells may be its one of the important mechanisms for multidrug resistance.
