摘要
目的建立测定小败毒膏中咖啡酸含量的高效液相色谱(HPLC)法。方法色谱柱为Agilent Technologies Zorbax Extend-C18柱(250mm×4.6mm,5μm),流动相为甲醇-0.01mol/L磷酸二氢钠溶液(23∶77),流速0.8mL/min,检测波长323nm,柱温40℃。结果咖啡酸质量浓度在1.00～40.00μg/mL范围内与峰面积线性关系良好(r=0.9996),平均加样回收率为97.85%,RSD=1.33%(n=6)。结论HPLC法简便易行,准确可靠,可用于小败毒膏的质量控制。
Objective To set up a method to determine the content of caffeic acid in Xiaobaidu Ointment.Methods The HPLC method was developed for the quantitative determination.The column：Agilent Technologies Zorbax Extend-C18 column（250 mm×4.6 mm,5 μm）.The mobile phase：methanol-0.01 mol/L sodium dihydrogen phosphate（23：77）.The flow rate was 0.8 mL/min,the detection wavelength was 323 nm and the column temperature was 40 ℃.Results The linear range for caffeic acid was 1.00-40.00 μg/mL,r=0.999 6.The average recovery rate was 97.85%,RSD was 1.33%（n=6）.Conclusion The method is simple,accurate and reliable,which can be used to control the quality of Xiaobaidu Ointment.
