摘要 目的探讨过氧化物酶增殖体激活受体-β(PPARβ-)在急性肺损伤(ALI)发生发展中可能的作用,以期从新的视角揭示ALI/ARDS的发病机理,并为ALI/ARDS的防治提供理论基础。方法采用颈静脉注射脂多糖(LPS)的方法复制大鼠ALI模型,随机分为对照组和LPS组。半定量RT-PCR方法测定肺组织中PPARβ-mRNA的表达水平;免疫组织化学染色法测定肺组织PPARβ-蛋白表达水平;同时测定动脉血气分析、肺组织湿/干(W/D)比值、肺组织MPO活性、光镜观察肺组织病理变化。结果LPS组大鼠PaO2显著降低(P<0.01),肺组织W/D比值、肺组织髓过氧化物酶(MPO)活性以及肺组织的病理积分均显著升高(P<0.05)。免疫组化显示对照组大鼠肺组织可表达PPARβ-蛋白,主要位于肺泡上皮细胞及肺间质细胞;与对照组相比,LPS组大鼠肺组织PPARβ-蛋白的表达均显著升高(P<0.05),分布在肺泡上皮细胞及炎性细胞。RT-PCR结果显示对照组肺组织表达一定量的PPARβ-mRNA,与对照组比较,LPS组大鼠肺组织PPARβ-mRNA的表达量显著升高(P<0.05)。结论正常大鼠肺泡上皮细胞及间质细胞存在PPARβ-,在ALI形成过程中PPARβ-蛋白和mRNA表达均显著增加,提示PPARβ-可能与ALI的炎症反应相关。 Objective To explore the effects of peroxisome proliferators-activated receptor-β(PPAR-β) in the pathogensis of acute lung injury(ALI) and provide the rationale for prevention and treatment of ALI/ARDS(acute respiratory distress syndrome).MethodsThe model of ALI rats was established with lipopolysaccharide(LPS,5 mg/kg).The rats were randomly divided into control group and LPS group.The semi-quantitative reverse transcription polymerase chain reaction(RT-PCR)technique was used to detect the expression of PPAR-β mRNA in the lung homogenate of rats.The immunohistochemistry(IHC) method was used for determining the protein expression of PPAR-β in the lung tissues of rats.The ratio of lung wet-dry weight(W/D),arterial blood gas analysis,the activity of myeloperoxidase(MPO) in lung homogenate and pathological change were observed respectively.Results The PaO2 of arterial blood gas analysis at all time in ALI group were significantly lower than those in control group(P0.01).The ratio of lung W/D and the level of pulmonary pathology accumulated points and MPO activation at 1h,2h,4h and 8h in ALI group were significantly higher than those in control group(P0.05).The results of immunohistochemistry(IHC) showed that the slight protein of PPAR-β was expressed in alveolar epithelial cells and lung interstitial substance cells in the control groups.relatively,the protein expression levels of PPAR-β increased significantly in the lung tissues of ALI rats(P0.05)and it was mostly expressed in alveolar epithelial cells and lung interstitial substance inflammatory cells.The results of RT-PCR showed the slight PPAR-β mRNA was expressed in the lung tissues of normal rats,relatively,the expression levels of PPAR-β mRNA increased significantly in the lung tissues of ALI rats(P0.05).Conclusions PPAR-β was expressed in alveolar epithelial cells and lung interstitial substance cells in the lung tissues of normal rats,the expression levels of PPAR-β mRNA and protein were increased remarkably in ALI group after LPS disposal,above results suggested PPAR-β alternation may be concerned with inflammation in ALI.
