胆道闭锁肝脏组织中SHH信号与上皮间充质转化关系的研究 被引量:11

Relationship of Sonic Hedgehog and Epithelium Mesenchymal Transition in the Development of Hepatic Fibrosis in Patients with Biliary Atresia

摘要 目的观察胆道闭锁(BA)肝纤维化发生过程中SHH信号与上皮间充质转化(EMT)过程的关系,探讨胆道闭锁肝纤维化过程中SHH信号在EMT过程中的作用。方法取经手术证实为BA的患儿(年龄1~3个月)肝脏组织10例,无消化道疾病新生儿(年龄1~3个月)尸检肝脏组织5例做正常对照,应用realtime RT-PCR及Western blot等方法检测SHH及反映EMT的指标E-cadherin、vimentin的表达水平。结果与正常对照组相比,BA患者肝脏组织中SHH表达水平明显升高,E-cadherin表达无明显变化,而vimentin表达水平明显增高。结论EMT在胆道闭锁肝纤维化中起重要作用,而SHH表达上调可能促进了这一过程,从而导致肝脏组织持续纤维化。 Objective To observe the relationship between sonic hedgehog(SHH)signaling and epithelium mesenchymal transition(EMT) and explore the regulating roles of SHH for EMT in hepatic fibrosis of biliary atresia.Methods Liver biopsy specimen from 10 infants(age 1 to 3 months)with biliary atresia and liver tissue from autopsy of 5 infant(age 1 to 3 months)without digestive disease were taken into study.Real-time RT-PCR and Western-blot were employed to detect the expression of SHH,E-cadherin and vimentin which represented the level of EMT.Results The expression of SHH in liver specimen of BA patients increased,the expression of E-cadherin was comparable to that of the control while Vimentin upregulated.Conclusions EMT procession was very important in the genesis of liver fibrosis in BA.Upregulation of SHH expression may act as an incentive to this procession.
