PTEN和Caspase-3在大肠癌中的表达及临床意义 被引量:19

The Expression and the Significance of PTEN and Caspase-3 in Colorectal Carcinoma

摘要 目的检测大肠癌组织中PTEN和Caspase-3的蛋白表达,了解与大肠癌临床病理的相关因素。方法采用S-P免疫组化法,检测PTEN和Caspase-3在各组织中的表达及意义。结果PTEN在正常大肠组织、大肠腺瘤、大肠癌组织中的表达率分别为100%、70%和51.61%;而癌旁组织的阳性表达率60.87%,与癌组织相似,两者有明显的同步表达性(P<0.01)。大肠癌中PTEN蛋白的阳性表达率明显低于非癌组(P<0.05)。Caspase-3在正常黏膜、大肠腺瘤及大肠癌中的阳性表达率分别为93.33%、60%、45.16%,在癌旁组织的表达率为52.17%。正常黏膜组织Caspase-3的表达高于大肠腺瘤和大肠癌(P<0.05)。PTEN表达随肿瘤分化程度的降低、临床分期提高而降低(P<0.05),Caspase-3阳性表达随肿瘤分化程度的降低而降低(P<0.05)。大肠癌组织中PTEN与Caspase-3的表达呈正相关(r=0.675,P<0.01)。结论PTEN、Caspase-3的表达下调与大肠癌的发生、发展有关,二者可作为判定大肠癌生物学行为和预后的临床参考指标。 Objective To determine the expression of PTEN and Caspase-3 in colorectal carcinoma and investigate the relationship between them and the clinicopathologic features.Methods The S-P immunohistochemical method was used to detect the expression and significance of PTEN and Caspase-3 in colorectal tissues.Results Expressions of PTEN in normal colorectal mucosa tissues,colorectal adenoma tissues and colorectal cancers are 100%、70% and 51.61%,respectively.Expression of PTEN in paracarcinoma mucosa is 60.87%,similar to that in colorectal cancer.Expression of PTEN in colorectal cancer is lower than that in non-cancer groups(P〈0.05).Expressions of Caspase-3 in normal colorectal mucosa tissues,colorectal adenoma tissues and colorectal cancers are 93.33%、60%、45.16%,respectively.And expression in paracarcinoma mucosa is 52.17%.Expression of Caspase-3 in normal colorectal mucosa tissue is higher than that in colorectal adenoma tissue and colorectal cancer(P〈0.05).The expressions of PTEN and Caspase-3 in well-differentiated adenocarcinoma were higher than that in poorly-differentiated adenocarcinoma(P〈0.05).Also,the lost or decreased expression of PTEN significantly correlated with clinicalpathological stage .Significant relationships were found between PTEN and Caspase-3(r =0.675,P〈0.01).Conclusion The experiment suggests that PTEN and Caspase-3 may play certain roles in the oncogenesis and progression of colorectal cancer.Their expressions may be favorable markers and useful prognostic indicators in the patients with colorectal cancer.
