VEGF在小剂量超短波促进种植骨髓间充质干细胞脱细胞支架修复大鼠坐骨神经损伤中的表达 被引量:3

The Expression of VEGF in the Low Dose Ultrashortwave Repairs Rats Sciatic Nerve Lesions by Using Acellular Allogenic Nerve Scaffold Implanted with Bone Mesenchymal Stem Cells

摘要 目的探讨血管内皮生长因子(VEGF)在小剂量超短波对种植骨髓间充质干细胞(BMSC)脱细胞支架修复大鼠坐骨神经损伤中的表达。方法将培养至第3代的大鼠BMSC注入已制备的脱细胞支架内,然后置于培养箱内联合培养3d,无菌条件下缝合到神经缺损处。实验鼠分为3组,即达尔伯克改良伊格尔培养基(DMEM)组、支架内注射BMSC组和注射BMSC后应用超短波(USW)治疗组;超短波治疗组在术后2d用小剂量超短波对移植动物的神经缺损处治疗;移植后第2,4,8和12周检测再生的神经中VEGF、S-100蛋白表达。结果各组VEGF在损伤后再生即有表达,并随时间延长呈上升趋势,在第4周达到顶峰,超短波治疗组表达高于其他组(P<0.05)。结论VEGF在超短波治疗早期的高水平表达可加速种植的BMSC分化或迁移的速度,并能促使再生神经周围的血供增加。 Objective To observe the expression of vascular endothelial growth factor(VEGF)in the low dose ultrashortwave repairs rats sciatic nerve lesions by using acellular allogenic nerve scaffold implanted with BMSC.Methods BMSC were injected into the prepared acellular nerve scaffold and maintained in DMEM complex culture medium for 72 h until the experiments were performed,then the graft sutured into defective sciatic nerve.Adult male Wistar rats(200~250 g)were divided into DMEM control group,BMSC-treated group and BMSC-treated ultrashortwave(USW)therapy group.USW groups began to therapy with the low dose ultrashortwave at 2 days after surgery.At 2,4,8 and 12 weeks after surgery,animals were sacrificed and took out of the regenerated nerves to detect the expression of VEGF and S100.Results The VEGF expression can be detected in regenerated nerves.The results displayed the positive expression of VEGF increased to peak 4 weeks after surgery,then decreasd gradually,meanwhile its expression were highest at same stage in USW group compaired with BMSC group and DMEM group.Conclusion Ultrashortwave therapy were able to promote or accelerate BMSC differentiate into Schwann cells or to promote Schwann cells'proliferation and migration.The high expression of VEGF at early stage of nerve repairing concluded that the low dose ultrashortwave was able to provide more blood supply for nerve regeneration.
