猪瘟胶体金快速诊断试纸条的研制 被引量:11

摘要 采用柠檬酸钠还原法制备胶体颗粒,用猪瘟(CSF)抗体标记纯化,并包被在玻璃纤维膜上,另外将纯化的CSF抗体和羊抗鼠抗体包被在硝酸纤维膜上,组装成CSF快速检测条,对猪瘟病毒进行检测.结果表明:使用所研制的CSF快速检测条检测收集的70份待检样品,共检出猪瘟阳性病例26例,而经病毒分离鉴定阳性病例有28例,阳性符合率达92.8%;将5份阳性猪瘟病毒稀释后用试纸条和ELISA方法进行敏感性实验,结果表明,诊断试纸条的敏感性较高,用试纸条对鸽新城疫病毒、鸡减蛋综合症病毒、猪传染性胃肠炎病毒、猪繁殖与呼吸障碍综合症病毒进行检测,无交叉反应. A method for determining swine fever virus(CSFV) by using colloidal gold immunochromatography assay(GICA)was developed by using the reducing method of citric acid sodium prepare colloidal particles,and putificating the colloidal gold particles with the CSF antibody.The gold-anti-CSFV were wrapped onto glass fiber membrance,and the purified CSF antibody and goat-anti-rat antibody were wrapped on nitrocellulose membrane to prepare a kit for CSF diagnosis.70 samples were detected for CSFV by the rapid diagnosis strip,of which 26 positive cases were detected and virus isolation and identification of CSFV indicated 28 cases as positive cases.The coincidence on positive rate of detection was 92.8 %.Five diluted positive CSFV samples were used to perform the sensitivity experiments with testing paper stripe and ELISA method,the results showed that the testing paper strip was with a higher sensitivity and had no cross reaction with ND,EDS-76,TGEV and PRRSV.It was confirmed that the diagnosis method was convenient,rapid and accurate,with high specificity,good repetition and without using any instrument.
