一株H5N2亚型鹦鹉源禽流感病毒的分离鉴定

摘要 在广东进行流行病学调查时从鹦鹉采集的拭子样本中分离到一株禽流感病毒,应用聚合酶链反应(PCR)、血凝抑制试验(HI)和基因测序等方法技术对其进行了亚型鉴定。结果表明,此分离株具有血凝活性,且被H5亚型禽流感病毒标准阳性血清特异性抑制;分别用针对禽流感病毒的M、H5亚型禽流感病毒的HA基因、N2亚型禽流感病毒的NA基因特异性鉴定引物对该分离株进行PCR扩增,均扩增出特异性目的片段;测序及BLAST分析表明其与H5N2亚型禽流感美国分离株的HA基因和NA基因核苷酸序列相似性高达97%以上,由此该分离株鉴定为H5N2亚型禽流感病毒,并命名为A/Parrot/Guangdong/268/2005。 An influenza virus strain was isolated from parrot in Guangdong.It was identified by PCR,hemagglutination inhibition and sequence analysis.These results showed that the isolate had hemagglutination activity and was inhabited by the standard antibody of H5 avian influenza virus;it was amplified by RT-PCR using the specific identification primers of M,HA and NA of avian influenza virus,and gained the specific segments.Compared with avian influenza viruses in GenBank,HA and NA genes of the isolate shared the closed similarities with the H5N2 avian influenza viruses from USA,the similarities were over 97%.Thus,the isolate was identified as a H5N2 avian influenza virus,named A/Parrot/Guangdong/268/2005.
