鸭梨叶片不定芽再生体系的建立 被引量:5

Establishment of shoot regeneration system from leaf of Yali pear

摘要 为建立鸭梨(Pyrus bretschneideri Rehd.cv.Yali)叶片不定芽再生体系,采用正交试验设计研究了不同培养基、植物激素和植物生长调节剂的种类及其浓度、蔗糖含量对鸭梨叶片不定芽再生的影响。结果表明,基本培养基是影响叶片不定芽的诱导、继代和生根的关键因素,分别以NN69、MS和1/2MS为宜;IBA是影响叶片不定芽增殖的重要因素;最适诱导培养基为NN69+BA3.0mg·L-1+IAA0.5mg·L-1,再生频率和再生芽数分别为80.00%和1.77;最适增殖培养基为MS+BA1.0mg·L-1+IBA0.5mg·L-1,增殖系数可达2.23;最适继代培养基为MS+BA1.0mg·L-1+NAA0.5mg·L-1,平均苗高4.98cm;最适生根培养基为1/2MS+蔗糖30g·L-1+IBA0.5mg·L-1,生根率和平均根数分别为50.00%和2.04。该再生体系可为鸭梨遗传转化提供有效的受体系统。 Effects of basal medium, the types and concentrations of plant hormones and plant growth regulators and content of sugar on the shoot regeneration from leaves of Yali pear (Pyrus bretschneideri Rebd. cv. Yali ) were studied using orthogonal experiment designs in order to establish shoot regeneration system from leaf of Yali pear. The results showed that the basal medium was the most important factor for inducing, subculture and rooting of the shoot from leaves and the suitable basal mediums were NN69, MS and 1/2MS, respectively. IBA played a vital part in proliferation of shoot. The optimum medium for shoot induction was NN69 +BA3.0 mg ·L^-1+IAA0.5 mg-L^-1, the regeneration rate and numbers of shoot were 80.00% and 1.77, respectively. The optimum medium for shoot proliferation was MS+BA 1.0 mg·L^-1IBA0.5 mg· L^-1 and the proliferation coefficient was 2.23. The optimum medium for shoot subculture was MS+BA1.0 mg· L^-1 +NAA0.5mg· L^-1 and the average height of shoot was 4.98 cm ; The optimum medium for shoot rooting was 1/2MS+sugar 30 g·L^-1IBA0.5 mg· L^-1 and the rooting rate and average number of rooting were 50.00% and 2.04, respectively. The regeneration system established in this paper could be used for genetic transformation in Yali pear.
