抗弓形虫IgM抗体时间分辨荧光免疫分析法的建立及初步应用 被引量:1

摘要 目的采用时间分辨荧光免疫分析法(TRFIA)建立高灵敏的抗弓形虫(TOX)IgM抗体的检测方法。方法采用二抗间接法建立抗TOX IgM抗体的TRFIA。结果方法的批内和批间变异系数(CV)分别为1.9%和3.8%,敏感性为0.4 U/mL,可测范围为0.4-200 U/mL,20%、50%和80%的有效剂量(ED20、ED50和ED80)分别为36.3、88.2和147.8 U/mL。检测健康体检人员的血清,标本的测定值为(5.3±2.3)U/mL,建议阳性阈值为10 U/mL。结论抗TOX IgM抗体的TRFIA敏感性高、稳定性好,在优生优育及器官移植上具有很好的应用前景。 Objective To develop a time-resolved fluoroimmunoassay(TRFIA) with high sensitivity for the detection of anti-Toxoplasma(TOX)-IgM antibody.Methods The tested specimen or reference standard was added to micro-titer plate coated with the TOX antigen and the europium-labelled anti human IgM antibody was added.Results The between-run and within-run coefficients of variation(CV) of this TRFIA were 1.9% and 3.8% respectively.The sensitivity was 0.4 U/mL.The range of testing was from 0.4 to 200 U/mL.The TRFIA provided a linear dose response with 20% effective dose(ED20),50% effective dose(ED50) and 80% effective dose(ED80) of 36.3,88.2 and 147.8 U/mL,respectively.The value of healthy subjects sera was(5.3±2.3) U/mL,so whose value over 10 U/mL would be reported as positive.Conclusions The TRFIA of anti-TOX-IgM antibody is a method with high sensitivity and stability and it has application perspective in aristogenesis and organ transplantation.
