大鼠新型隐球菌脑感染的模型建立 被引量:1

Establishments of rat model with cerebral cryptococcus neoformans infection.

摘要 目的探讨免疫抑制状态下大鼠侧脑室注射新型隐球菌菌悬液较理想的浓度和剂量,建立稳定的大鼠脑感染模型。方法将40只免疫抑制的Wistar大鼠分成A、B两组,分别向颅内注射0.1ml和0.05ml新型隐球菌菌悬液。每组再分为4个亚组,每亚组5只,分别注射1×103cfu/ml、1×104cfu/ml、1×105cfu/ml、1×106cfu/ml浓度的菌悬液,每日观察。未死亡大鼠在7d、14d、21d、28d行MRI扫描,全部实验结束后处死大鼠观察病变处病理变化。结果(1)一般情况:1×103cfu/ml和1×104cfu/ml注射浓度时两组未见大鼠死亡。(2)MRI表现:大鼠7d时可见脑膜轻度线样强化,14d时见脑实质内感染病灶,21d时可见脑室扩大。(3)病理检查:脑膜及脑实质内病变处可见隐球菌菌体,局部见淋巴细胞浸润。结论免疫抑制状态下在大鼠侧脑室内注射浓度为1×104cfu/ml,剂量为0.1ml新型隐球菌菌悬液,可以建立比较稳定的大鼠脑感染模型,并用于新型隐球菌脑感染的MRI研究。 Objective To investigate the ideal concentration and dose of intracerebral injection suspension with cryptococcus neoformans and then establish the stable infected rat model.Methods 40 immunosuppressive Wistar rats were divided into A and B groups and injected with cryptococcus neoformans of 0.1ml and 0.05ml doses;each group was divided into 4 subgroups,and each subgroup had 5 rats.Every subgroup were injected with suspension of cryptococcus neoformans of 1 × 103、1 × 104,、1 × 105、1 × 106 cfu/ml concentration respectively.Rats were observed everyday.The survival rats were scanned by MR on the 7th、14th、21st、28th day.Rats were executed to observe the pathological changes of lesions at the end of all tests.Results(1) All the rats injected with concentration of 1 × 103 and 1 × 104cfu/ml were alive.(2) Imaging findings:on the 7th day,rats had mild meningeal enhancement;on the 14th day,rats had parenchyma lesions,on the 21st day,rats were seen ventricular dilatation.(3)Pathological examination:cryptococcus neoformans and lymphocytic infiltration were seen at meningeal and brain parenchymal lesions.Conclusion Intracerebroventricular injection of suspension of cryptococcus neoformans with concentration of 1 × 104 cfu/ml and dose of 0.1 ml can establish the stable cerebral infected rat model.The rat model are suitable for MRI study to cerebral cryptococcus neoformans infection.
