摘要
通过RT-PCR,从椰心叶甲蛹总RNA中扩增2个与酚氧化酶基因相关的DNA片段,经克隆,测序得到2个片段的碱基序列,分子量分别为417bp和638bp。同源性比对表明,417bp和638bp片段与黄粉甲原酚氧化酶同属一类;417bp和638bp的cDNA序列比对发现,638bp片段内包含了417bp片段的全序列,同时还存在2个插入序列,这2个插入序列基本都处于编码酚氧化酶氨基酸序列的阅读框之外。
Two phenoloxidase cDNA fragments of 417 bp and 638 bp were amplified from total RNA of pupae of coconut leaf beetle (Brontispa longissima) by using RT-PCR strategy with a pair of degenerated primers and cloned successfully. Sequence analysis showed that these two fragments had high identity of deduced amino acids with the known prophenoloxidase cloned from Tenebrio molitor. Alignment of these two fragments showed that the 638 bp fragment contained the whole sequence of the 417 bp fragment and had two inset sequences which generally fall out of the open reading frames of an amino acid sequence encoding prophenoloxidase.
