摘要
目的利用表面增强激光解析离子化飞行时间质谱(SELDI-TOF-MS)技术绘制一氧化碳(CO)中毒后不同恢复程度的患者血清中差异蛋白质峰的表达谱,建立一种方便临床快速、敏感、高效的预后评估新技术。方法收集60例急性CO中毒患者的血清样本,按预后程度不同分为痊愈、植物状态和死亡3大类别,利用弱阳离子芯片CM10和SELDI-TOF-MS技术分析不同类别患者血清中的差异表达蛋白质峰,联合应用明显差异表达的蛋白质峰建立分类决策树,随机选取30例样本对决策树进行盲法检验,得出决策树模型的分类准确率。结果共获得149个有效蛋白质峰,利用m/z=5 770、m/z=5 849和m/z=7 791的3个显著差异表达的蛋白质峰成功建立分类决策树,准确率为96.7%(48/50),敏感性为90.0%。结论SELDI-TOF-MS技术可较快速且准确地评估急性CO中毒患者的预后,将为今后临床评估提供新的技术支持。
Objective To search for specific protein biomarkers in the serum of different prognosis patients with acute carbon monoxide poisoning by surface enhanced laser desorption-ionization time-of-flight mass spectrometry (SELDI-TOF-MS) ,and to find a new technology for fast,sensitive and efficient prognosis evaluation. Methods Serum samples were obtained from 60 patients with acute carbon monoxide poisoning, who were classified into recovery, vegetative state,and death. Protein chip CM10 and SELDI were used to detect differential expression proteins,and the classification decision tree was established via combined protein peaks. The accuracy of classification decision tree was assessed by 30 samples which were randomly selected. Results 149 protein peaks were detected,and the classification decision tree was established via three protein peaks of m/z= 5 770, m/z= 5 849 and m/z= 7 791. The accuracy rate was 96.7%(48/50),sensitivity was 90.0%. Conclusion SELDi-TOF-MS technique may provide a fast and efficient prognosis evaluation for patients with acute carbon monoxide poisoning,and give a new reference for clinical prognosis evaluation in the future.
