摘要
用肠炎、鼠伤寒沙门氏菌鞭毛蛋白分别免疫56日龄ICR小鼠,经间接ELISA和直接凝集试验测定证实.产生了血清分型H抗原的抗体,亦产生了针对鞭毛蛋白共同抗原组分的抗体。同时,用灭活全菌抗原免疫3日龄伊莎鸡,经单抗CB8+de7竞争ELISA测定,灭活肠炎、鼠伤寒沙门氏菌免疫组同样测出较高滴度共同抗原表位的抗体。进一步用都柏林、鼠伤寒和肠炎沙门氏菌活菌分别人工感染56日龄ICR小鼠、3日龄伊莎鸡和140日龄SPF鸡.用单抗竞争ELISA亦检测出鞭毛蛋白共同抗原表位的抗体。这些结果表明:沙门氏菌鞭毛蛋白共同抗原表位可作为新的检测对象,在该菌感染诊断方面具有重要意义。研究中证实.单抗竞争ELISA为检测该类表位抗体提供了新方法。
Mice were immunized at 56-day of age with purified flagellin Hi or H(g,m) from S. typhimurium and S. enteritidis, respectively. Flagellins could elicit both serotype-specific H antibody with indirect ELISA and DA which titers were higher than 1: 10 000 and 1: 1 000, respectively, and the antisera of common epitopes on flagellins with indirect ELISA which titers were 1: 000~1: 1 000, in contrast, these serum samples did not react with S. pullorum either in indirect ELISA or in DA. Meanwhile, sera from chickens immunized with killed S. enteritidis or S. typhimurium contained the specific antibodies against common epitopes of Salmonella flagellins, while sera from those chickens inoculated with killed S. pullorum showed no reactivity with flagellins in the competitive ELISA, which is based on monoclonal antibodies (MAbs) CB8 and de7 against the generically-specific common epitopes on Salmonella flagellins and purified flagellins. In further study, 56-day mice, 3-day Salmonella-free chickens and 140-day SPF chickens were inoculated orally with live S. dublin, S. typhimurium and S. enteritidis, respectively. After two weeks of final inoculation, these animals produced the specific antibodies against those common epitopes detected by competitive ELISA. These results showed that the generically-specific common epitopes on Salmonella flagellins could be used as a detectable marker in the process of Salmonella infections, and suggested that the corresponding MAbs-based competitive ELISA was very useful in the diagnosis of Salmonella infections.
